一款新设备能从唾液拭子中提取人的DNA,只需几分钟就能提供全部的分析数据和基因组测序数据。

美国华盛顿州西雅图的华盛顿大学和NanoFacture公司(Bellevue, WA, USA)的科研人员开发的这种新设备用显微探针浸入唾液、痰液或血液等液体样本,并给液体加上电场,吸引粒子在探针表面周围聚集。较大的粒子碰到针尖并转向,但和DNA差不多大的分子粘到探针上,通过毛细管作用被表面捕获。用这项技术分离和提纯DNA只需两三分钟。

这款手持式设备一次能清理四份人体液样本,但这项技术经过扩展后一次可制备96份样本,这是大规模处理的标准规格。华盛顿大学的工程师们还利用同样的探针技术设计了一种像铅笔一样大的设备,病人可以戴着它回家,也可以把它发给海外服役的军人。病人可以刮拭面颊,收集唾液样本,并在现场处理DNA,然后送回医院和化验室分析。

首席研究员、华盛顿大学机械工程系副教授Jae-Hyun Chung博士说:“提取DNA非常复杂。目前的过程就像用起重机收集人的头发。”

传统方法用离心机旋转并分离DNA分子,或用微型滤器从液体样本中拉出DNA分子,但这些过程需要20到30分钟才能完成,可能还需要用到过多的有毒化学品。医院和研究实验室用新设备更容易从人体液样本中分离DNA,从而有助于基因组测序、疾病诊断和法医调查。
